Understanding which proteins interact in a cell is a fundamental question of biochemistry. As almost all cellular processes are performed by ‘molecular machines’, reliable protein interaction data can be gleaned by isolating and dissecting them. The primary goal of this proposal is to unravel the protein interaction network for the E. coli inner membrane by characterizing the oligomeric protein complexes contained in it. The data obtained from this study will be used to define biochemical pathways, and functions for membrane proteins, and soluble proteins associated with the membrane. As 40-50% of all E. coli proteins are anticipated to function at the inner membrane, this study should contribute a significant volume of novel information. To date, protein interaction maps for membrane proteomes remain largely unchartered and this study will be the first of its kind.
