The innate immune system provides a fast response against invasion by pathogens and also determines the quality of the adaptive immune responses. Chlamydia, Mycobacterium and Rotavirus species are intracellular pathogens that cause a variety of severe human diseases. Because of the high incidence of these infections world wide, new or improved tools are necessary to reduce the burden that these pathogens pose on public health. We plan to investigate the role of innate immunity in resistance and pathogenesis of Chlamydia, Mycobacterium and Rotavirus infections in patients and animal models. Our long-term goal is to provide, by targeting the innate immune system, a novel vaccine and therapeutic strategy. Specifically we will examine the roles of a) pathogen recognition by innate immune responses b) cytokine modulation of innate responses in the outcome of infection with each of the three pathogens. We will also focus on the identification of human genes responsible for predisposition to disease. Finally, we intend to use innate immune molecules such as cytokines as adjuvants to improve vaccine strategies.
