By activating three receptor subtypes, galanin has been shown to participate in regulation of several physiological mechanisms. The present research project will focus on the development of pharmacological tools necessary to understand the extended role of the galaninergic system within the nervous system behind the disorders such as depression, drug (nicotine) addiction and feeding behaviour. Recent knowledge about the ligand binding rules and molecular properties of the galanin receptor subtypes will be explored in order to further understand the requirements for development of subtype selective ligands. Modulation of the receptor expression in cellular and animal tissue by antisense and siRNA oligonucleotides targeting the specific receptors will further help us to understand the role of the different galanin receptors. The developed pharmacological tools will subsequently be utilized in behavioural models of depression, drug (nicotine) addiction and feeding behaviour to specifically understand the role of the galanin receptors within these important physiological conditions.
